| Abstract: | The present invention relates to a i.e. 6-thioguanine encapsulated albumin-gold nanocluster composition, the process of making the same, and therapeutic use thereof, for the treatment of cancer. The synthesis of Albumin-Au-TG comprises a one-pot green approach where an aqueous HAuCl4 or NaAuCl4 solution is stirred with bovine or human serum albumin (BSA or HSA) and 6-TG dissolved in NaOH for 12 h at 37°C, followed by addition of ethanol to precipitate the drug loaded nanoparticles. Detailed characterization studies indicate that the loading of hydrophobic 6-thioguanine in Albumin-Au platform happens through Au-S, amide, and -S- S- bond formation. Albumin-Au-TG shows red fluorescence in solution which become helpful to illustrate the drug loading, sustained drug release behaviour and intracellular uptake of the nanocluster composition. The MTT assay on cancer cells shows significant cell death after 72 hrs of treatment. The current formulation offers improved solubility, circulation lifetime, bioavailability, site specific transport of purine drug molecules due to nanometric dimension, specific targeting of diseased cells and reduced off-target toxicity. |
