Abstract: The present invention discloses DNA vaccination with plasmid encoding the cysteine protease C (cpc) gene of Leishmania donovani to induce effective protection against visceral leishmaniasis (VL). The DNA vaccine entrapped in the cationic liposomes comprises of a plasmid vector pVAX1 encoding L. donovani cpc gene operably linked to transcriptional regulatory sequences. The invention relates to efficacy of liposomal formulation containing the bilayer-associated adjuvant MPLA, encapsulating the DNA construct pVAX1-cpc, against lethal L. donovani infection model in BALB/c mice. This formulation of liposomal DNA vaccine is highly effective in inducing robust cell-mediated immune response against leishmanial parasites upon intramuscular administration.
