Abstract:
The invention unveils the synergistic potential of linalool and linalyl acetate combination in the treatment of psoriasis like skin conditions. The invention reveals the role of novel composition comprising linalool and linalyl acetate in the alleviation of level of Th-17 cell producing cytokines through IL-23/Th-17axis. Furthermore, the invention discloses the activity of linalool and linalyl acetate combination in the reduction of Psoriasis Area Severity Index (PASI) score. Specifically, the invention unveils the synergistic activity of linalool and linalyl acetate combination in the alleviation of the level of biochemical parameters like Th-17 cell expressing cytokines (IL-22 and IL-17) and Th-1 cell cytokines (IL-1ß and TNF-a) as compared to linalool and linalyl acetate alone at the same dose. More specifically, the invention unveils the activity of linalool and linalyl acetate combination in the restoration of histopathological changes such as dermal hyperplasia and hyperkeratosis toward normal skin conditions as evidenced by histopathological studies. Even more specifically, the invention discloses the development of molecular docking simulation models revealing binding mode conformations of linalool and linalyl acetate on selected human psoriasis targets/receptors such as IL-17 (PDB: 3JVF), IL-22 (PDB: 3G9V), IL-23 (PDB: 3DUH), TLR-8 (PDB: 3W3J), IL-17a (PDB: 2VXS), IL-17b (PDB: 2VBC), IL-17f (PDB: 1JPY), IL-22R-a-1 (PDB: 3DLQ) and IL-22R-a-2 (PDB: 3G9V).
