| Abstract: | The present invention relates to a synergistic antihypertensive herbal formulation consisting of four components that are ar-turmerone, curcumin, demethoxycurcumin, and bisdemethoxycurcumin with level of all curcuminoids not less than 16%. The formulation is effective against L-NAME and SHRs model of hypertension in improving level of DHEA and hemodynamics. Herbal formulation is effective in reversing the down and up regulated mRNA expression of DHEA biosynthesis pathway in L-NAME and SHRs model of hypertension. Down regulated protein expression of CYP11A1 and CYP17A1 of DHEA biosynthesis pathway in liver tissue of L-NAME and SHRs models of hypertension were restored by the proposed formulation. This formulation is safe in acute and sub-acute oral toxicity in mice model. |
