Abstract: The invention provides novel halogen rich salicylanilides compounds that enhancing histone acetylation. An efficient process was developed for the synthesis of novel  HAT activators. The disclosed compounds increased cell permeability without conjugation to the carrier, enhanced in vitro acetyltransferase activity and activation at  lower concentration in both in vitro and in vivo makes it a better small molecule activator of p300 acetyltransferase than its parent molecule TTK21. The invention  further provides methods for treating neurodegenerative disorders, cancer and spinal cord injury.
