Abstract:
The present invention relates to a technology platform with a novel aptamer and antibody combination, for detection of SARS-CoV-2 viral protein. It relates to the generation of an aptamer which is a single stranded DNA sequence that can selectively and sensitively bind to the spike protein of SARS-CoV-2. The novel aptamer was generated using a biolayer interferometry based invitro protocol which was further subjected to in silico maturation using genetic algorithm and docking studies to select the final sequence. The champion aptamer could specifically bind to SARS-CoV-2 viral protein in a concentration dependent manner and also demonstrated ACE2 inhibition which provided preliminary evidence of its selectivity. This novel aptamer was further combined with monoclonal anti-SARS CoV-2 antibody as conjugating reagent for the development of a lateral flow assay based rapid antigen test (RAT).
