| Abstract: | The present invention aims to develop highly efficient lipid-based, drug/gene delivery nano-formulations using glycolipids (GL) and phospholipids (PL) isolated from crude rice bran gum obtained from rice bran oil refinery. Present invention shows that formulations carrying cationic lipid such as D1 with or without PL and cholesterol showed greater penetration through the blood-brain-barrier compared to standard DOPC : Chol formulation. Further supplementation of minute quantity of GL to each formulation enhanced the permeation and retention of respective nano formulations in mice brains. Moreover, formulations carrying both GL & PL and with or without D1 showed efficient gene transfection in brain cancer cells. Additionally, GL/PL carrying formulations with or without D1 additionally encapsulating hydrophobic anticancer drug, docetaxel showed efficient antitumor effect. Replacing synthetic lipids with natural mixtures of GL/PL reduces cost of production, increases biological output as well as provide value-addition to rice bran gums, helping in overall sustainability of the industry. |
