Abstract:
Mitigating the multifactorial progression of cancer demands a multi-targeted therapeutic strategy. Although FDA-approved PARP inhibitors have shown clinical  success, their efficacy is largely restricted to BRCA-mutated cancers. Emerging evidence suggests that mTOR inhibition can sensitize cancer cells to PARP inhibition.  In light of this, we aimed to develop a first-in-class dual PARP–mTOR inhibitor capable of targeting both BRCA-deficient and BRCA-proficient cancers, thereby  expanding the therapeutic horizon for a broader population of cancer patients. The invention relates to purine-based phthalazinones as dual PARP [Poly (ADP-ribose)  polymerase] and mammalian Target of Rapamycin (m-TOR) inhibitors, process for the preparation and uses thereof as potential anticancer agents.
