Abstract:
The present invention relates to noncytotoxic selective mTORC1 inhibitors for chronic liver diseases like obesity, non-alcoholic fatty liver disease (NAFLD), and hepatocellular carcinoma and process for preparing the same. The synthesized compounds are capable of selectively inhibiting the mTORC1 pathway and decreasing the total lipid accumulation both in cultured HepG2 cell lines as well as mouse primary hepatocytes. The mTOR inhibitory efficacies were also observed in vivo using C57Bl/6 mice models. Moreover, the compounds demonstrate no significant cytotoxicities, and therefore offer viable means for designing noncytotoxic selective mTORC1 inhibitors useful in the treatment of chronic liver diseases and metabolic syndromes.
