| Abstract: | Trypsin/trypsin-like proteases have been reported to promote the entry of SARS-CoV-2 into host cells. The spike has a protease cleavage site between the S1 and S2 domains. The cleavage properties of proteases can be used to design drug screening assays for screening antiviral drug candidates for spike cleavage. In the claimed invention we developed a proof of concept detection system for screening drugs against proteases that cleave spikes between S1 and S2. We have designed fusion substrate proteins comprising a reporter protein, a protease cleavage site between S1 and S2, and a cellulose binding domain. Due to the presence of Cellulose Binding Domains (CBDs), the substrate protein may be immobilized on cellulose. When the protease cleaves the substrate, the CBD remains bound to the cellulose and the reporter protein is removed. The released reporter protein can be used as a readout of protease activity. | ||
