Abstract:
RNA-based fluorescent probes are currently limited by their low selectivity toward RNA versus DNA, and low specificity to different RNA structures. Poor membrane permeability is another defect of existing fluorogenic RNA probes for intracellular imaging. In this work, novel Indole based probe Compound 2, was developed for the rapid and selective detection of intracellular rRNA (rRNA). An appropriate lipophilic balance with the hydrophilic amine group and hydrophobic Indole in probe, as well as its high-water solubility, makes the molecule suitable for bioimaging of rRNA. Furthermore, high contrast and brightness imaging in live cells as well as the first successful example of effective viscosity selective light-up probe for distinct nucleolus imaging in live cells. These results are particularly critical for a better insight of nucleolar dynamic mechanisms of a change in RNA content and distribution, and localization throughout a cell cycle. The present invention provides a new tool that allows imaging of RNA in live or fixed cells with high Stokes shift and specificity, which will be helpful to explore the intracellular RNA concentration changes in the diseased conditions such as in highly proliferating cancer cells and neurodegenerative Alzheimer disease. This can be further used to develop suitable to develop diagnostic kit as for understanding changes in environment of nucleoli with respect to rRNA in real time.
