Abstract:
The present invention relates to the preparation of 8-phenyl-purine-6-amino compounds with general structure I in free form or in acceptable salt form to  identify small molecule activator of mTOR, which can be used to augment the effector function (IFN? production) of T cells and hence heightened their antitumor  response. More particularly, the invention relates to small molecules where R1, R2 and R3 are as defined in the description, capable of IFN? production via mTOR  activation pathway to elicit durable antitumor repose in the host. The invention describes strategies to generate T cells with high effector function, yet in a relatively  short period of time so that a robust and durable anti-tumor response can be achieved in the host.
