| Abstract: | The present invention discloses a novel therapeutic strategy that combines the CDK inhibitor, IIIM-368, with established cytotoxic anti-cancer agents to target a broad spectrum of human cancer cell lines. The combination showed moderate to additive effects for most cancers. However, a strong synergy was observed between IIIM-368 and Paclitaxel in lung cancer cells, allowing reduced Paclitaxel and IIIM-368 dosage while maintaining efficacy. This treatment is safe for normal cells supported by high selectivity index. Mechanistically, the combination of IIIM-368 and Paclitaxel induces cell cycle arrest in the S and G2 phase of the cell cycle and induces apoptosis in A549 cells. Importantly, the combination exhibits antitumor efficacy in A549 human xenografts in nude mice, resulting in significant tumor size reduction. and demonstrates efficacy against Paclitaxel-resistant A549 cells, suggesting its potential to overcome drug resistance mechanisms. This synergistic combination offers a promising new therapy for non-small cell lung cancers. | ||
